Mango-3 WT FL Aptamer is a compact 45-nucleotide fluorogenic RNA aptamer that binds the thiazole orange derivative TO1-Biotin with high, low-nanomolar affinity and switches on its fluorescence by more than 1,000-fold, producing a bright green signal (emission ~535 nm) with an exceptionally high fluorescence quantum yield (~0.55). Fluorescence activation occurs because the aptamer folds into a compact G-quadruplex that, together with an embedded non-canonical duplex, clamps the otherwise non-fluorescent dye into a rigid, planar conformation. This makes Mango-III one of the brightest light-up aptamers and a powerful tool for visualizing RNA directly in biological systems without the need for protein-based tags.
Unlike traditional fluorescent reporter systems that rely on bulky protein fusions, Mango-3 is a small, genetically encodable RNA tag that can be expressed inside living cells, enabling real-time tracking of RNA molecules — including short, structured non-coding RNAs — with minimal structural disruption.
