
E coli elongator tRNA coding for Methionine CAT anti-codon
E. coli tRNA-Met (CAT), elongator is a 77-nucleotide transfer RNA that delivers methionine to the ribosomal A site during chain elongation. E. coli carries two functionally distinct classes of methionine tRNA: the initiator tRNA-fMet, which is formylated and used to start translation, and the elongator tRNA-Met supplied here, which is never formylated and functions only in elongation. Although both share the CAU anticodon, they are distinguished at the top of the acceptor stem, the initiator carries a C1·A72 mismatch required for recognition by methionyl-tRNA formyltransferase, whereas the elongator has a standard Watson-Crick base pair and is therefore excluded from initiation and handed instead to EF-Tu. This product is the wild-type sequence encoded at the metU locus (identical to metT), supplied as an unmodified in vitro transcript with an intact 3′ CCA end, ready for aminoacylation by methionyl-tRNA synthetase or by flexizyme.
